According to his aunt who was there for his birth and named him Willie Hugh, no. . . the claim is that he was actually born a few minutes before midnight on April 29, 1933. If, in fact, his actual DOB is 4/29/33, how cool is that? 4 plus 29 equals 33! Happy day, Willie Nelson!

So, is it his birthday or not? According to birth records, yes.
